Brain-penetration plays a crucial role for compounds directed to treat CNS diseases. Dennis will present state-of-the-art knowledge of in silico, in vitro, and in vivo tools to assess, predict, and optimize brain-partitioning for CNS drugs. He’ll explain different multiparameter scores to pre-select compounds for synthesis and discuss how they perform against machine-learning models. Furthermore, he’ll share which in vitro assays should be used to decide on advancing CNS active compounds. The determination of unbound brain concentration, why it matters and how it can be optimized will be discussed. Dennis will address some common misconceptions in CNS drug discovery, and will conclude with a case study from a CNS drug discovery campaign in Human African Trypanosomiasis. Dennis Koester, Ph.D. is an Associate Director in the Medicinal Chemistry Group at FibroGen in San Francisco, CA. He earned his PhD from Göttingen University with Prof. Daniel Werz working in carbohydrate chemistry. During his graduate work he pursued an interdoc at Kyoto University with Prof. Yoshiaki Nakao in cooperative catalysis. He completed two Postdocs at Münster University with Prof. Frank Glorius working on Rh catalysis, and at Stanford University with Prof. Barry Trost in enantioselective and transition metal-catalysis.
